Vasoactive Intestinal Peptide (VIP) is a 28-amino acid neuropeptide (MW ~3,326 Da) belonging to the glucagon/secretin superfamily. Originally isolated from porcine intestinal extracts by Said and Mutt in 1970, VIP is widely distributed throughout the central and peripheral nervous systems, gastrointestinal tract, and immune tissues.
Research investigations have documented VIP's potent anti-inflammatory and immunomodulatory properties across diverse experimental systems. Studies have examined its effects on Th1/Th2 immune balance, dendritic cell tolerogenesis, and regulatory T-cell induction. Preclinical research in models of autoimmune and inflammatory conditions has investigated VIP-mediated suppression of pro-inflammatory cytokines (TNF-alpha, IL-6, IL-12) and enhancement of anti-inflammatory mediators (IL-10). Investigators have also explored VIP's neuroprotective properties, effects on circadian rhythm regulation via the suprachiasmatic nucleus, and role in pulmonary vascular biology and airway smooth muscle relaxation.
Mechanistically, VIP signals through two class B G-protein-coupled receptors: VPAC1 (widely distributed) and VPAC2 (concentrated in CNS and immune tissues). Receptor activation triggers cAMP/PKA signaling cascades that modulate NF-kB transcriptional activity, CREB-dependent gene expression, and downstream inflammatory mediator production.
