DSIP (Delta Sleep Inducing Peptide) is a neuropeptide with the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu and a molecular weight of approximately 848 Da. First isolated from cerebral venous blood of rabbits during electrically induced sleep by Schoenenberger and Monnier in 1977, DSIP has been identified in multiple brain regions and peripheral tissues including the gut and adrenal glands.

Research investigations have examined DSIP's effects on sleep architecture, stress response modulation, and neuroendocrine signaling. Preclinical studies have documented alterations in sleep EEG patterns, particularly increased delta wave activity during slow-wave sleep phases. Investigators have explored its potential interactions with the hypothalamic-pituitary-adrenal (HPA) axis, including effects on ACTH and cortisol signaling under stress conditions. Additional research has examined DSIP's role in circadian rhythm modulation, pain perception pathways, and oxidative stress responses. Studies have also investigated its effects on LH and GH release patterns in animal models.

The mechanism of action remains an active area of investigation. Proposed pathways include modulation of GABAergic neurotransmission, interaction with opioid receptor systems (particularly delta receptors), and effects on glutamatergic NMDA receptor sensitivity. DSIP may also function as a stress-protective factor through modulation of free radical metabolism.
